Development of a technique to study gene function during regeneration in an adult newt. We have identified several interesting candidate genes that are highly upregulated during newt limb and spinal cord regeneration but are limited to testing only a few of these genes for in vivo function, because there are no current universal methods for studying gene function in the adult newt. One of the projects in my laboratory is to develop such a method. We have preliminary data suggesting that morpholinos might be able to knockdown gene expression in the adult newt, and we are currently continuing to investigate this method for testing gene function. Other possible methods we may test include RNA interference and local transgenesis using recombinant lentiviruses. 12/2008 |
