A study of a zebrafish line, TC-8, proven to develop T cell malignancies which most closely resemble human T cell lymphoblastic lymphoma. This line was identified from a ‘mutagenesis screen,’ where hundreds of different fish carrying a GFP transgene (causing their T cells to be green) were exposed to a chemical teratogen, and their progeny screened for abnormal T cell phenotypes. Several areas of investigation with the TC-8 line are underway, including: (1) phenotypic characterization of the disease using immunohistochemical, RNA in situ, and RT-PCR approaches, (2) establishing conditions for in vitro culture of malignant cells, and (3) efforts to map the mutated gene underlying these malignancies. Again, students involved in this project will have the freedom to concentrate their efforts in whatever area(s) they find most interesting. 1/2008 |
